PROBLEM TO BE SOLVED: To provide improved regulatory elements that enable high expression and sustained expression in applications such as gene therapy.SOLUTION: The present invention provides hybrid regulatory regions using elements from one or more ubiquitin promoters together with one or more viral promoters such as viral promoters from cytomegalovirus (CMV). The present invention further provides DNA vectors that provide high expression and persistent expression of associated coding sequences. Particularly, the use of CpG-reduced plasmid vectors containing a CMV-Ub hybrid promoter provides long-term expression and efficacy required for a practical gene therapy.SELECTED DRAWING: None |
