Fluorescent imaging with substituted cyanine dyes

Compounds and methods are disclosed that are useful for noninvasive imaging in the near-infrared spectral range. The cyanine compounds of Formula I are presented:;whereinQ is a portion of a polymethine bridge:;
Also included are bioconjugates of the compounds of Formula I, methods of labeling biomolecules with the compounds, and methods of imaging.

A near-infrared (NIR) compound of the formula: wherein R1 is LYZ; L is an optional member selected from the group consisting of a bond, a C1-C20 alkylene, and a C1-C20 alkenylene; wherein the alkylene or alkenylene is optionally interrupted by at least one heteroatom; Y is an optional member selected from the group consisting of a bond, —O—, —S—, —NH—, —NHC(O)—, —C(O)NH—, —NR15—, —NR15C(O)—, —C(O)NR15—, —N(Z)—, —N(Z)C(O)—, and —C(O)N(Z)—; R15 is a member selected from the group consisting of alkyl and alkoxycarbonylalkyl, wherein the alkyl is optionally interrupted by at least one heteroatom; Z is R16; or alternatively, —Y—Z is a member selected from the group consisting of —N(Z)2, —N(Z)C(O)Z, and —C(O)N(Z)2, and the two Z groups may optionally be linked to form a cycloalkynyl group; and R16 is member selected from the group consisting of azido, alkynyl, a pegylated azido, a pegylated alkynyl, a pegylated cycloalkynyl, a pegylated spirocycloalkynyl, an o -diarylphosphino aryl ester, and an ortho substituted phosphine oxide aryl ester, DBCO, DBCO-1, TPPME, BARAC, DIFO, BCN, DIBO, TMDIBO and DIFO3, wherein the compound has emission in the near-infrared.
