The present invention relates to a vaccine composition comprising an MTBK 20620 protein. According to the present invention, in the case of using the MTBK 20620 protein as an antigen, the composition exhibits high IFN-γ production capacity in the spleen and the lung of an animal model with the infection induced by tubercular bacillus, and is capable of effectively preventing tuberculosis by maintaining the formation of acquired rapid and protective immunity and the memorization of T cells. In addition, according to the present invention, the MTBK 20620 protein can prevent the reinfection or the recurrence of highly pathogenic tuberculosis with a high recurrence rate by being used for treating the tuberculosis, and can be efficiently used for increasing treatment efficiency.
