PROBLEM TO BE SOLVED: To provide an antibody comprising a polypeptide having a variant Fc region with increased binding to FcRn, the antibody having a low rate of clearance in blood circulation.SOLUTION: There is provided a method for making a phage library including a group of variants from the human antibody Fc gene and performing screening using the binding to FcRn as an indicator. By the method, Fc variants exhibiting high binding to FcRn, particularly amino acid modification sites thereof, are determined, and anti-CD20 antibody human IgG is reconstructed using the Fc variants. The anti-CD20 antibody human IgG exhibits ADCC activity to chronic lymphocytic leukaemia cells. |
