The present invention relates to a rice straw saccharification method using pre-treatment both radiation and alkali, and to a lipid production method of microalgae using the same. The rice straw saccharification method using pre-treatment both radiation and alkali can be helpfully used in producing lipid using the microalgae, by increasing a cellulose content when treating both a large amount of gamma rays and the alkali, and increasing decomposable properties of rice straw which removes a large amount of lignin and hemicellulose, increasing a glucose and a xylose yield in enzyme hydrolysis, and confirming that Chlorella protothecoides, which are ipid-producing microalgae, produce lipid when culturing a rice straw hydrolyzate as a carbon source.
