PROBLEM TO BE SOLVED: To provide a method and a composition in particle-based detection of a target molecule using a linking molecule.SOLUTION: A method of generating a complex comprising a nucleic acid target molecule is provided which comprises: mixing in a reaction mixture a substrate comprising a plurality of capture probes bound to the substrate, a sample suspected of containing a nucleic acid target molecule, and a linking molecule. The linking molecule is a target-specific binding portion which specifically binds to an existing nucleic acid target molecule, and when the linking molecule includes a plurality of target-specific binding portions, each of the target-specific binding portions includes one or a plurality of target-specific binding portions which specifically bind to different regions of the existing nucleic acid target molecule, and one or a plurality of capture probes specific binding portions which specifically bind to at least one of a plurality of capture probes. |
