Improvements in and relating to the manufacture of pyrimidine derivatives

Novel compounds of the general formula wherein R represents a straight-chain or branched alkylene radical and R1 represents a hydrogen atom or the acyl radical of an aliphatic carboxylic acid containing 1 to 6 carbon atoms or of an aromatic or araliphatic carboxylic acid, are prepared by reacting a hydroxy alkoxy-acetal of general formula HO-R11-O-CH2-CH(OY)2 or a 2-alkoxy or 2-halogen-1,4-dioxacycloalkane or a 1,4-dioxa-2-cycloalkene, where R11 is a saturated straight chain or branched alkylene radical and Y represents an alkyl radical containing 1 to 6 carbon atoms with an N,N-disubstituted formamide and an inorganic acid chloride, e.g. PCl5, POCl3 or COCl2 or in the case of 1,4-dioxa-2-cycloalkenes adding an orthoformic acid ester to the double bond in the presence of an acid catalyst, e.g. BF3, and then condensing the product with guanidine. Alternatively, 2 - amino - 5 - iodo - pyrimidine can be reacted with an alkali metal alcoholate MO-R-OH, in which M is the alkali metal cation, in the presence of copper or a copper compound. Another method of preparation involves reacting a compound of the general Formula II, wherein R1, R2, R3 and R4 are each hydrogen atoms or alkyl radicals containing 1 to 4 carbon atoms, and R5 represents a saturated or unsaturated, straight-chain or branched or cyclic or araliphatic hydrocarbon radical, in a non-oxidizing strong inorganic acid, e.g. phosphoric acid with an alkali metal iodide, preferably at temperatures between 80 DEG and 150 DEG C. When R1 in Formula I represents a hydrogen atom, the free hydroxy group may be esterified with a carboxylic acid, and when R1 is an organic acid radical, the ester group may be hydrolysed and/or converted into a salt.
