Disclosed are a method for preparing an amylose magnetic beads and a method for purifying protein using the same. The method for purifying target protein using the amylose magnetic beads according to the present invention comprises the steps of: (a) reacting one or more substrates, selected among glucose, oligosaccharide and sucrose, and magnetic nanoparticles which are guest molecules to prepare amylose magnetic bead composites; (b) binding the target protein with the amylose magnetic bead composites; and (c) separating and purifying the amylose magnetic bead composites to which the target protein is bound.
