PROBLEM TO BE SOLVED: To find a molecular target specific to a human leukemia stem cell (LSC), and provide treatment means or the like leading to complete cure of acute myeloid leukemia (AML).SOLUTION: There are provided: a testing method of predicting first occurrence or recurrence of AML includes (1) a process of measuring an expression level of an LSC marker gene in a biological sample collected from a subject regarding a transcription product or a translation product of the gene, and (2) a process of comparing an expression level obtained from the measurement process with a reference value, in which the LSC marker gene is composed of 2 to 218 genes; a therapeutic agent of AML targeting an LSC containing a substance which can suppress expression of a gene selected from the LSC marker gene or activity of a translation product of the gene, as an active ingredient; and a manufacturing method of a sample containing hematopoietic cells for autologous or allograft transplant to an AML patient in which the method includes a process of obtaining a sample from which the LSC was purged by using at least one kind of LSC marker as an indicator.
