Provided are an extract isolation apparatus and a method of operation thereof; the extract isolation apparatus comprises an adsorption mechanism (11), a separation disc (12), a pipetting mechanism (13), an injection mechanism (14), and a motion mechanism (15); the separation disc is a rotatable annular mechanism, and along same is provided a plurality of reaction cuvette positions (121) spaced at intervals; along the rotational path of the separation disc are distributed a plurality of pipetting positions and at least one injection position; the adsorption mechanism is also an annular mechanism; the adsorption mechanism is provided with a plurality of adsorptive components (111) along its perimeter, and said adsorption mechanism is at least provided with one vacant position (112) having no adsorptive component; the motion mechanism drives the separation disc and adsorption mechanism to rotate and stop independently, and the vacant position moves with the position requiring no adsorptive force. The separation disc and adsorption mechanism are driven to rotate and stop independently, such that the adsorptive mechanism can provide an adsorptive force or not provide an adsorptive force, according to requirements.
