The present invention relates to a method of quantitatively detect thrombin using an enzyme reaction, a kit to detect thrombin and a manufacturing method thereof and, more specifically, relates to a method of detecting thrombin electrochemically using fibrinogen tagged with ferrocene, a thrombin detection kit to perform the method thereof, and a manufacturing method of the thrombin detection kit. The kit to detect thrombin and the method to quantitatively detect thrombin using the same of the present invention presents a new technology of detecting thrombin without immobilizing protein on a surface; thus accurately and rapidly detecting a target protein without having a problem of reducing a protein activity.
