PROBLEM TO BE SOLVED: To provide a detection method of wheat, using a wheat-detecting PCR primer having specificity to the wheat, capable of widely detecting wheat at a high sensitivity (50-500 fg wheat DNA in a reaction system of 25μl PCR reaction liquid) and obtaining no target amplified product from cereals other than wheat, such as barley and rye, and to provide a primer set of the method. SOLUTION: The detection method of wheat includes a step for obtaining amplified products by performing PCR which uses a primer having a specific base sequence and a primer set comprising the primer having the specific base sequence, and a step for judging the existence of wheat by using the amplified products as the index. In addition, the detection method of wheat includes a step for obtaining amplified products by performing PCR which uses a primer set composed of a mixed primer by four kinds of primers having specific base sequences, and a step for judging the existence of wheat by using the amplified products as the index.
