Method for preparing three-dimensional tissue structures (A) that comprises first growing cells (isolated from cartilage, bone, bone marrow aspirate, nucleus pulposus or anulus fibrosus) on a solid carrier in an adhesion medium. Method for preparing three-dimensional tissue structures (A) comprises: (A) growing cells (isolated from cartilage, bone, bone marrow aspirate, nucleus pulposus or anulus fibrosus) on a solid carrier in an adhesion medium; (B) maturing the grown cells for 2-5 days with periodic change of medium; (C) rinsing the cell layer at least once with cell culture buffer devoid of calcium and magnesium ions; (D) incubation for 30-90 minutes in similar buffer, in an incubator, to separate the cell layer from the carrier; (E) transferring the released cell layer to cell culture medium and mechanical separation into cell heaps; (F) incubating the heaps for up to 7 days, with periodic change of medium, so that a cell cluster matures; and (G) subjecting the cluster cyclically to at least one pressure load and subsequent mechanical shaping. An independent claim is also included for three-dimensional structures produced by the new process. |
