An engine oil consumption measuring system and method of a vehicle are provided to measure the oil consumption at low cost with simple method by measuring the voltage of level sensor of the measuring unit connected to the engine when the oil pressure of the engine and the measuring unit are the same. An engine oil consumption measuring system of a vehicle comprises an oil pan(30) and an engine(40) connected to an oil supply system(10) via a valve block(21) and a coupling(24); a measuring unit(50) which is connected to the engine and the valve block and equipped with a level sensor(51) measuring the mass of oil; and temperature sensors(63,65) installed in the oil pan and the measuring unit, respectively. The engine oil consumption measuring system measures the voltage change of the level sensor when supplying a constant amount of oil to the engine through the oil supply system and determines based on the relation between the measured voltage and the oil level of the measuring unit or the oil amount of the oil pan.
