The invention relates to inflammatory bowel disease and particularly to methods and compositions for screening for, diagnosing the presence or progression of, or detecting the risk of developing ulcerative colitis and/or colonic Crohn's disease in a subject. In one embodiment, the invention relates to a method of screening for, diagnosing the presence of, or detecting a risk of developing, ulcerative colitis comprising detecting the presence of at least one disease associated PTPRS gene variant in a sample of a subject wherein the presence of the disease associated PTPRS gene variant is indicative of ulcerative colitis or an increased risk of developing ulcerative colitis compared to a negative subject not having the disease associated gene variant of PTPRS.
