<p>Promoter-reporter gene fusion comprising of 1f-aromatase promoter according to SEQ ID NO: 1, sequence is available in electronic form and may be found at ftp://ftp.wipo.int/pub/published pct sequences/publication, and reporter gene, is new. Independent claims are: (1) a method of determining of a tissue-selective action of substances that bind to the androgen receptor, comprising: (a) determining of the modulation of aromatase regulation using the promoter-reporter gene fusion in a reporter gene assay in cell culture systems for identifying both agonistic and antagonistic activities; (b) determining of the modulation of another androgen-regulated promoter; and (c) comparing (a) and (b), to achieve a complete in vitro characterization of substances with respect to selective aromatase modulation on the if-aromatase promoter or in direct comparison with other aromatase-regulated promoters; and (2) in vitro test system for the identification and characterization of substances that bring about an increase in aromatase expression selectively in the central nervous system and/or are suitable for exerting a tissue-selective influence on the activity of the aromatase, where the test system contains the fusion of reporter gene and aromatase promoter.</p> |
