Gene expression profiling from FFPE sample

Provided is a method of synthesizing cDNA from the polyadenylated RNA from cells of an formalin-fixed and paraffin-embedded (FFPE) sample comprising:a) extracting said RNA from said cells; b) heating said RNA extracted from said cells at about 70°C for greater than 60 minutes; and c) contacting said extracted RNA with a primer comprising an oligo dT sequence under conditions that result in the synthesis of a first cDNA strand complementary to said extracted RNA. Further provided are variants of the method using different primers, RNA extraction methods and cDNA synthesis.
