PURPOSE: A biosensor for detecting very small amount of sample and manufacturing method thereof are provided to connect nano-electrode through enzyme-metal ion precipitation and enhance the conductivity. CONSTITUTION: A method for manufacturing a biosensor for detecting very small amount of sample comprises: a step of fixing a receptor molecule which selectively binds to a target material in an insulated nano-electrode chip; a step of binding an enzyme (31) to the fixed receptor; a step of treating metal ion in the enzyme and precipitating a metal ion on nano-electrode surface through enzyme precipitation; and a step of washing and drying.
