PROBLEM TO BE SOLVED: To precisely estimate a staining quantity of a staining sample dyed by a predetermined dyeing pigment. SOLUTION: In one of the embodiments, a storage 16 stores a plurality of reference spectra in different conditions of a staining pigment staining the staining sample of an object to be observed as reference spectrum information 163. A spectrum acquiring part 141 obtains spectral characteristic values each pixel of staining sample images with staining samples picked up. A sample preparing condition estimating part 142 estimates a preparation condition when, for instance, the staining sample is prepared, and obtains the preparation condition of the staining sample. A reference spectrum determining part 146 selects the reference spectrum corresponding to the preparation condition of the staining sample from the reference spectrum information 163 and determines the optimal reference spectrum of the staining pigment. A pigment quantity estimating part 147 estimates the pigment quantity of staining pigment staining the staining sample using the optimal reference spectrum of the staining pigment based on the spectral characteristic values obtained by the spectrum acquiring part 141.
