PROBLEM TO BE SOLVED: To provide compositions and methods for producing translational components that expand the number of genetically encoded amino acids in vertebrate cells.SOLUTION: A tRNA transcription cassette comprises flanking human tRNA-Tyr sequences, human tRNA-Tyr, and one or more kinds of amber-suppressing Escherichia coli tRNATyr. The human tRNA-Tyr is upstream of the amber-suppressing Escherichia coli tRNATyr. The tRNA transcription cassette comprises, in order from the 5' side to the 3' side, (a) a 5' flanking human tRNA-Tyr sequence, human tRNA-Tyr, and a 3' flanking human tRNA-Tyr sequence, and (b) a 5' flanking human tRNA-Tyr sequence, amber-suppressing Escherichia coli tRNATyr, and a 3' flanking human tRNA-Tyr sequence. |
