The present invention relates to a pharmaceutical composition comprising an NDRG3 expression or activity inhibitor as an active ingredient for preventing and treating inflammatory diseases. Specifically, NDRG3 mediated by lactate which is generated by a hypoxia reaction promotes expression of cytokine which mediates an inflammatory reaction through a lactic acid-NDRG3-Raf-ERK signal path, thereby usefully using the inhibitor for inhibiting the NDRG3 expression or activity as a pharmaceutical composition for preventing and treating inflammatory diseases.
