At least one light source is provided to emit a switching radiation for switching a first subset in the sample and to emit an excitation radiation for exciting the first subset. At least one of the light sources is arranged to radiate through the sample, and at least either of switching and fluorescence excitation of UF in the sample is carried out in at least one direction approximately perpendicular to an optical axis and in the focus of the objective lens O.
