PROBLEM TO BE SOLVED: To provide a microscope for imaging structures of samples with high spatial resolution.SOLUTION: A microscope includes a diffraction-limited resolution volume with a plurality of dye molecules UF which can be switched between different states, where at least one state is fluorescent, the fluorescence being collected by an objective lens O and imaged on a spatially resolving detector by an optical system, and have a distribution density that is greater than the inverse of the diffraction-limited resolution volume. At least one light source is provided to emit a switching radiation for switching a first subset in the sample and to emit an excitation radiation for exciting the first subset. At least one of the light sources is arranged to radiate through the sample, and at least either of switching and fluorescence excitation of UF in the sample is carried out in at least one direction approximately perpendicular to an optical axis and in the focus of the objective lens O. |
