A PIK3CA gene mutation detection kit can be used for detecting PIK3CA gene mutation associated with cancers. The kit comprises: (1) an internal reference detection reagent which comprises internal reference gene specific primers, an internal reference gene specific probe, and a dNTP solution; (2) a PIK3CA mutation detection reagent which comprises PIK3CA gene mutation type specific primers, a PIK3CA gene mutation type specific probe, internal control gene specific primers, an internal control gene specific probe, and a dNTP solution; (3) Taq DNA polymerases; and (4) a PIK3CA positive quality controller.
