POLYPEPTIDES AND BIOSYNTHETIC PATHWAYS FOR THE PRODUCTION OF STEREOISOMERS OF MONATIN AND THEIR PRECURSORS

PROBLEM TO BE SOLVED: To provide a method for synthesizing monatin through the production of indole-3-pyruvate (I3 P) from L-tryptophan.SOLUTION: The method comprises the steps of: producing a monatin precursor (MP) including at least R monatin precursor (R-MP) from I3 P with enzymes selected from L-tryptophan aminotransferase, L-aromatic aminotransferase, L-aspartate aminotransferase or L-amino acid oxidase and a combination thereof, and then producing monatin from MP with one or more enzymes selected from D-aminotransferase, D-amino acid dehydrogenase and a combination thereof. In some embodiments, the method produces monatin or certain stereo isomers of monatin such as R,R monatin and S,R monatin and salts thereof by using polypeptides and biosynthetic pathways.
