摘要 Disclosed are methods for the fabrication of non-spherical nano/micro-particles with controlled size, shape, surface properties, and mechanisl stiffness over a wide size range. Also disclosed are methods for using these particles as systemic (i.e., intravascular) delivery system for delivery of imaging and/or therapeutic agents to selected cells and/or tissues of the mammalian body. In certain applications, the dislosed particles are discoidal, with a circular or rectangular base; have a characteristic size of ~1,000 nm, and are composed of varying combinations of poly(lactic acid co-glycolic acid) (PLGA) and polyethylene glycol (PEG). Also disclosed are methods for unprecedented tumor accumulation (over 10% ID/g tumor, without molecular or magnetic targeting) of the particles, and their use as in vivo contrast enhancements either for PET- or magnetic resonance-based imaging modalities. Certain of the disclosed particles can also be employed for thermal ablation therapy, and/or magnetic dragging/steering.
