血友病Ａモデルブタの作出

摘要

PROBLEM TO BE SOLVED: To provide a hemophilia A model pig presenting a clinical state of hemophilia and a spontaneous bleeding symptom, useful in developing an effective treatment method.

SOLUTION: The method of creating the hemophilia A model pig includes: (a) a step for making a targeting vector aiming at an exon 16 of the VIII factor gene; (b) a step for introducing the targeting vector produced in the step (a) and selecting a gene-recombined cell generated from the homologous recombination; (c) a step for removing a nucleus from a collected egg matured inside the body of a pig; (d) a step for injecting a nucleus of the gene-recombined cell selected in the step (c) into the egg whose nucleus is removed in the step (c); and (e) a step for subjecting the egg into which the nucleus is injected in the step (d) to an activation process and transplanting a nucleus-transplanted embryo after the activation process to an oviduct or uterus of a female pig.
