PROBLEM TO BE SOLVED: To provide a method for inducing chromatin into pronuclei by using a cell-free reconstitution system that uses mammalian oocyte extract.SOLUTION: This invention relates to a method for inducing pronuclei from chromatin, comprising the following steps (a)-(c): (a) a step 1 of mixing chromatin and a transparent fraction of an egg extract prepared from activated eggs derived from a mammal, and incubating this mixture; (b) a step 2 of furthermore adding an egg extract or opaque fraction to the mixture obtained after the step 1, and incubating the resultant mixture; and (c) a step 3 of furthermore adding an egg extract or opaque fraction to the mixture obtained after the step 2, incubating the resultant mixture, and confirming whether or not the chromatin in the mixture has differentiated into pronuclei. |
