PROBLEM TO BE SOLVED: To provide compounds and compositions comprising the compounds useful in the treatment of cancer.SOLUTION: Compounds and compositions comprising the compounds useful in the treatment of cancer are described herein. The compounds and compositions can be used to modulate an isocitrate dehydrogenase (IDH) mutant (e.g., IDHIm or IDH2m) having alpha hydroxyl neoactivity. A method of treating a cancer characterized by having an IDH mutation comprises administering to a subject a therapeutically effective amount of a specific compound or a pharmaceutically acceptable salt thereof.
