The present invention relates to a yeast strain having a double reporter system for a novel two-hybrid system and, more specifically, to a yeast strain comprising a c-Myc reporter expression cassette and a Cre reporter expression cassette at the same time, and a library-library protein interaction detection method using the yeast strain. A double reporter yeast strain according to the present invention, when applied to protein interaction detection, provides a means for effectively processing massive information about protein binding pairs at a library level.
