<p>PROBLEM TO BE SOLVED: To provide a method for introducing nucleic acid into Botryococcus braunii cells, Botryococcus braunii cells to which nucleic acid has been introduced by the method, a method for producing Botryococcus braunii cells maintaining the nucleic acid in the genome and a method for producing hydrocarbon using the cell line.SOLUTION: Studying the techniques to introduce nucleic acid into the cells of Botryococcus braunii by removing the outer coat barrier (extracellular matrix) formed by Botryococcus braunii around the cells, as a result of trial-and-error, the inventors have found that efficient nucleic acid introduction can be achieved by isolating a population of cells lacking the outer coat that appear at a low frequency in the culture process, and then carrying out electroporation to the obtained population.</p> |
