摘要 The present invention provides a method for determining the 3-dimensional or 3-D atomic resolution structure of a biomolecule using chemical shift data obtained from the biomolecule having one or more selectively isotopically labeled biomolecule monomers and interrogation of same using an NMR device. Methods also comprise use of a low-field NMR device and structure determination method to determine the 2-D and 3-D structure of the biomolecule, for example, a polynucleotide.

A method for determining the 2-D or 3-D atomic resolution structure of a polynucleotide, the method comprising: (a) providing a polynucleotide sample comprising a polynucleotide, the polynucleotide comprising at least one nucleotide isotopically labeled with one or more atomic labels selected from the group consisting of 2H, 13C, 15N, 19F and 31P; (b) obtaining a NMR spectrum of the polynucleotide sample using a NMR device; (c) determining a chemical shift of the one or more atomic labels; and (d) determining a 2-D or a 3-D atomic resolution structure of the polynucleotide from the chemical shifts determined in step (c).
