A process for preparing concentrated polyunsaturated fatty acid oil, which comprises: subjecting oil and fat that contain polyunsaturated fatty acid as a fatty acid composing the oil and fat to an alcoholysis reaction using lipase in the presence of alcohol, added water and at least one compound, as a reaction additive, selected from magnesium oxide, magnesium hydroxide, calcium oxide and calcium hydroxide; and separating a glyceride fraction that contains concentrated polyunsaturated fatty acid, wherein the amount of the reaction additive is 0.05% to 5% (w/w) relative to the oil and fat containing polyunsaturated fatty acid, and the amount of the added water is 5% to 20% (v/v) relative to the alcohol, wherein the polyunsaturated fatty acid is eicosapentaenoic acid, docosahexaenoic acid, arachidonic acid, or a combination thereof, and wherein the lipase is selected from the lipases obtained from the microorganisms of Alcaligenes sp., Thermomyces lanuginosus, Burkholderia cepacia, and Pseudomonas fluorescens.
