The present invention provides a method for manufacturing a reaction mixture of 10-hydroxystearic acid having high yield through stirring vegetable oil containing oleic acid and oleic acid hydratase manufactured by using biotransformation under temperature and pH conditions and a method for isolating and purifying 10-hydroxystearic acid having high purity by neutralizing, dehydrating and drying processes, and adding an acidic solution and obtaining 10-hydroxystearic acid through neutralizing and washing processes after removing other fatty acids and oleic acid which do not react by adding an alkaline solution in the form of a salt.
