PROBLEM TO BE SOLVED: To provide a method for selectively recognizing base pair in DNA sequence by a polypeptide, modified polypeptides which specifically recognize one or more base pairs in a DNA sequence, DNA which is modified so that it can be specifically recognized by a polypeptide, uses of the polypeptide and DNA in targeting specific DNA, and a method for modulating expression of target genes in a cell.SOLUTION: There is provided a method for producing a polypeptide that selectively recognizes at least one base pair in a target DNA sequence. The method comprises synthesizing a polypeptide comprising repeat domain. The repeat domain comprises at least one repeat unit derived from a transcription activator like (TAL) effector. The repeat unit comprises a hypervariable region that determines recognition of a base pair in the target DNA sequence. The repeat unit is responsible for the recognition of one base pair in the DNA sequence.
