PROBLEM TO BE SOLVED: To provide a positioning device of a glass plate and a processing method of a glass plate which can improve processing productivity of the glass plate by protecting a glass plate pressing part in polishing the glass plate.SOLUTION: A positioning mechanism 41 included in a positioning device of a glass plate G includes a glass plate pressing part 42 which presses an outer circumference edge of the glass plate G, and a drive mechanism 43 which drives the glass plate pressing part 42. The drive mechanism 43 includes a vertical drive part 44 which moves the glass plate pressing part 42 vertically, and a press driving part 45 which moves the glass plate pressing part 42 to a pressing direction for pressing the glass plate G. The vertical drive part 44 is structured so as to move the glass plate pressing part 42 downward from the glass plate G. The press driving part 45 is structured so as to move the glass plate pressing part 42 to an inner side S1 from an outer circumference end surface Ga of the glass plate G.
