A glucose response promoter is provided to induce restoration of physiological properties of insulin-producing beta cell, and control glucose homeostasis in a patient suffering from diabetes by inducing insulin expression as increase of glucose concentration, so that the promoter is useful for treatment of diabetes. A glucose response promoter specific to the isolated tissue contains polymerase binding domain 3' comprising a liver pyruvate kinase(LPK) promoter to at least one tripartite transcription factor binding cis element having a hepatocyte nuclear factor-1(HNF-1) element, CAAT/enhancer binding protein(C/EBP) response element and glucose-response element(GRE). A host cell comprises the glucose responsive promoter. The diabetes are prevented and treated by administering an effective amount of a virus particle having a vector containing the glucose responsive promoter comprising a polymerase binding domain 3' to at least one tripartite transcription factor binding cis element having the hepatocyte nuclear factor-1 element, a C AAT/enhancer binding protein response element and the glucose-response element which are operationally linked to a nucleic acid encoding insulin of which expression is tissue specific and glucose responsive to a patient suffering from diabetes. Further, the HNF-1 element comprises 25 to 30 nucleotide, and the GRE comprises 45 to 50 nucleotide.
