摘要 A method for immobilizing a protein to a substrate by using light irradiation is provided to improve immobilization and orientation properties of protein on the substrate through a covalent bond, inhibit release from the substrate, and uniformly orient the protein on the substrate, so that the method is applicable to biosensor and bioreactor. A protein with reactive phenyldiazirine represented by the formula(1) contains an antibody capable of recognizing a light cross-linker group selected from FERM BP-10762, FERM BP-10763, FERM BP-10764, FERM BP-10825 and FERM BP-10826, wherein R1 is hydrogen atom or optionally substituted alkyl group; R2 is hydrogen atom, halogen atom, alkoxy group or alkyl group optionally substituted by alkylene oxide; and the antibody has the amino acid sequence containing (a) SEQ ID NOs:1, 2 and 3, (b) SEQ ID NOs:4, 5 and 6, (c) SEQ ID NOs:7, 8 and 9, and (d) SEQ ID NOs:10, 11 and 12. A method for immobilizing a protein to a substrate comprises the steps of: (1) forming a light cross-linker containing reactive phenyldiazirine derivatives to the substrate surface; (2) reacting the protein with the cross-linker on the substrate surface to immobilize it to the substrate; and (3) irradiating a light to the substrate to form a cross-linked structure between the substrate and protein. Further, the antibody is selected from a group consisting of a chimeric antibody, a complementary determining region-transplanting antibody, a single-stranded antibody and an antibody fragments thereof.
