The invention provides for a random insertional mutagensis and screening method for the reliable and efficient identification and preparation of functional protein variants which have an improved phenotype when compared to the protein from which they are derived. The method involved the random insertion of a polyamino acid into a selected protein scaffold, to prepare a library of protein variants, and selection of those variants which present the desired functional characterisitics, such as solubility and/or the ability to trigger an immunogenic response.
