NOVEL PHYSIOLOGICALLY ACTIVE POLYPEPTIDE

PURPOSE:To provide a novel polypeptide having a specific amino acid sequence and having an antitumor activity of as high as about 2.8 times that of human TNF protein. CONSTITUTION:Human TNF gene-expression plasmid (preferably pTNF 416) is cut with a proper restriction enzyme and a specific region is removed from the cut fragments. The gene is restored by the use of a synthetic oligonucleotide having a proper base sequence to obtain the polypeptide of formula. The polypeptide is preferably the one having Met bonded to the amino terminal. The objective polypeptide having antitumor activity can be produced in high efficiency, by transforming a microorganism with a recombinant plasmid containing a DNA region coding the above polypeptides.
