PURPOSE:To determine the titled compound useful for diagnosing liver and biliary system without pretreatment of a test specimen, by adding 3-oxo-5beta- steroidDELTA<4>-dehydrogenase to the test specimen in the presence of a reducing coloring agent and measuring absorbance of formed colored substance. CONSTITUTION:3-Oxo-5beta-steroidDELTA<4>-dehydrogenase is added to a test specimen in the presence of a reducing coloring agent (e.g. nitro blue tetrazolium, etc.), accurately reacted at 37 deg.C for 10min, a reaction terminator (e.g. 0.1N-HCl, etc.) and adsorbance of formed colored substance at 540nm wavelength is measured to determine 3-oxo-5beta-steroid. Serum, plasma, urine, etc., may be used as the test specimen.
