PROBLEM TO BE SOLVED: To provide a method for determining gain of a transmission detector in an imaging type microscope apparatus capable of performing observation while making a time when a sample is exposed to light as short as possible. SOLUTION: When observation of the sample using fluorescence is finished, an image is formed by using observation light from the sample without changing the state of the apparatus. When brightness of the image at such a time is defined as T, and light quantity of a light source 111 is defined as L, the gain G of the transmission detector 113 is obtained as (1) G=T/(L×α). Provided thatαis a constant determined in advance by an optical element existing between the objective lenses 118a to 118c of an illuminator and the transmission detector 113.
