<P>PROBLEM TO BE SOLVED: To provide a method for synthesizing a single strand DNA having a desired chain length at a desired position in a template nucleic acid. <P>SOLUTION: This method for synthesizing the nucleic acid comprises (1) a process for annealing a primer having a complementary sequence in a nucleotide chain on the 3'-terminal side of a target nucleic acid sequence to be synthesized, to a template nucleic acid containing the target nucleic acid sequence and used as a starting point for an elongation reaction of the target nucleic acid sequence, and a blocker having a sequence complementary to an upstream side nucleotide sequence in the just front of the 5'-terminal of the target nucleic acid sequence and used for terminating the elongation reaction, and (2) a process for synthesizing the complementary chain of the target nucleic acid sequence from the primer as a starting point with a nucleic acid synthetase substantially not exhibiting a 5'→3' exonuclease activity, after the above-described process (1). <P>COPYRIGHT: (C)2005,JPO&NCIPI |
