(i) reacting a sample with a cholesterol determination enzyme (e.g., a cholesterol ester hydrolase) in a reaction solution containing a surfactant capable of predominantly inhibiting the reaction between the cholesterol determination enzyme and sdLDL-C, thereby removing HDL-C, VLDL-C, CM-C and LgLDL-C from the sample; (ii) adding a reagent capable of inducing the reaction of sdLDL-C remaining in the reaction solution after the step (i) to produce hydrogen peroxide or reduced coenzyme, and then measuring hydrogen peroxide or reduced coenzyme produced; and (iii) determining the sdLDL-C level in the sample based on the measurement value obtained in the step (ii) and a calibration curve that has been prepared previously.
