<p>It is intended to provide a vector DNA for amplification outside the chromosome of a host cell and a method for amplifying a target gene using the vector. The vector according to the invention comprises a mammalian replication initiation region which functions in a eukaryotic cell and a nuclear matrix association region and is characterized in that it is linear and at least one end is in the form of a hairpin structure. By using the vector of the invention, a target gene can be highly amplified in the DM form, i.e., extrachromosomally. Accordingly, it is considered that a level of a protein expressed from the target gene also increases in proportion to an increase in the number of amplified copies of the target gene without suffering repetitive sequence-dependent transcriptional repression.</p> |
