A process for producing 7-methoxy-3-desacetylcefalotin by using a bio catalyst is provided to obtain a cefoxitin synthetic intermediate through hydrolysis process of being performed in water in the presence of a catalyst consisting of enzymes. A process for producing 7-methoxy-3-desacetylcefalotin of the formula (I) comprises a step of hydrolyzing an acetyl group of a compound of the chemical formula (II). The hydrolysis is performed in water in the presence of a catalyst consisting of at least one enzyme having hydrolastic acetyl activity at a temperature of -10-45 and in pH 5-9. The catalyst is made of the fixed enzyme on the solid support. The enzyme is obtained from microorganisms selected from the group consisting of Rhodosporidium toruloides, Bacillus pumilus, Escherichia coli, Acremonium chrysogenum and Streptomyces clavuligerus.
