<p>Purifying an analyte secreted by cells comprises incubating the cells in a culture medium supplemented with a serum substitute, separating a supernatant and producing a retentate by ultrafiltration, incubating the retentate with functionalized magnetic beads in a buffer solution, and magnetically separating the beads. The serum substitute is derived from a natural serum and contains no substances with a molecular weight of 30 kD or less, and the magnetic beads are functionalized with at least 10-generation dendrimers with modified termini. Independent claims are also included for: (1) serum purified to remove low molecular weight components; (2) functionalized magnetic beads produced by reacting magnetic beads having surface amino groups with methyl acrylate in an organic solvent, separating the beads and washing them with an organic solvent, reacting the beads with ethylenediamine in an organic solvent, separating the beads and washing them with an organic solvent, repeating the above steps n times (n is preferably 0-9), suspending the beads in a solution containing a functionalizing substance, separating and washing the beads and suspending the beads in water or buffer solution.</p> |
