PURPOSE: A primer pair for amplifying microsatellite DNA marker is provided to accurately and simply distinguish tuna from swordfish. CONSTITUTION: A primer pair for amplifying a microsatellite DNA marker to determine tuna species from swordfish comprise: a primer pair with sequences of sequence numbers 1 and 2; a primer pair with sequences of sequence numbers 3 and 4; primer pair with sequences of sequence numbers 5 and 6; a primer pair with sequences of sequence numbers 7 and 8; a primer pair with sequences of sequence numbers 9 and 10; a prime pair with sequences of sequence numbers 11 and 12; or combination thereof. A method for distinguishing tuna species from swordfish comprises: a step of extracting DNA from the sample; a step of preparing PCR solution containing the extracted sample DNA and primer pair; a step of performing PCR; and a step of analyzing genotype of PCR product.
