PROBLEM TO BE SOLVED: To provide human antibodies bound to human insulin-like growth factor-I receptor (IGF-IR), derivatives of the antibodies (Fabs, single chain antibodies, bi-specific antibodies, or fusion proteins), and to provide therapeutic and diagnostic methods of using the antibodies and derivatives. SOLUTION: The invention relates to isolated human antibodies or fragments thereof which bind specifically to the human insulin-like growth factor-I receptor (IGF-IR), and also have at least one property selected from (i) inhibiting binding of IGF-I or IGF-II to IGF-II to IGF-IR; (ii) neutralizing activation of IGF-IR by IGF-I or IGF-II; (iii) reducing IGF-IR surface receptor by at least about 80%; and (iv) binding to IGF-IR with a Kd of about 1×10-10M-1or less.
