In vitro method of screening candidate compounds for the prevention and/or treatment of an infection caused by a bacterium of the genus Mycobacterium, comprises providing a candidate compound, and determining whether the candidate compound inhibits the efflux pump CTPC and/or the efflux pump CTPG of a bacterium of the genus Mycobacterium, where the candidate compound which inhibits the efflux pump CTPC is identified as the candidate compound. Independent claims are included for: (1) an inhibitor of an efflux pump CTPC of a bacterium of the genus Mycobacteriumor inhibitor of an efflux pump CTPG of a bacterium of the genus Mycobacteriumcomprising anti-CtpC antibody, antisense oligonucleotides targeting the gene CtpC, ribozymes directed against mRNA of CtpC and aptamers, anti-CtpG, antisense oligonucleotides targeting the gene CtpG, or ribozymes directed against mRNA of CtpG and aptamers, for use as a medicament in the treatment or prevention of infection by a bacterium of the genus Mycobacterium; (2) a recombinant bacterium of the genus Mycobacterium, comprising at least one mutation of CtpC gene, which is either inactivation or deletion of all or part of the gene CtpC producing defect of synthesis of the efflux pump CtpC or a mutation that leads to the synthesis of efflux pump CTPC unable to export the zinc and/or at least one mutation of the gene CtpG, which is either inactivation or deletion of all or part of the gene CtpG producing defect of synthesis of the efflux pump CtpG, or a mutation that leads to the synthesis of an efflux pump CtpG unable to export zinc; and (3) an immunogenic or vaccine composition comprising the bacterium and an excipient.
