Disclosed herein are benzoquinoline (tetraneazine) compounds of the formula (I), wherein R1-R27 are independently selected from the group consisting of hydrogen and deuterium, and at least one of R1-R27 is deuterium. Additionally, a pharmaceutical composition comprising a compound of the formula (I), for the treatment of a VMAT2-mediated disorder, such as the chronic hyperkinetic movement disorder, Huntington's disease. A pharmaceutical composition comprising a compound of formula (I) and an additional therapeutic agent selected from the group consisting of olanzapine and pimozide, or the group consisting of benzodiazepines and antipsychotics.
